Although there have been major advances in science and technology in healthcare, scientists are still working aggressively to curb the burden of cancer on patients and the healthcare system at large. A vital role of chemokines is their ability to guide the migration of cells in response to an opportunity (food/glucose/energy) or a problem (e.g., cancer). Chemokines are fundamentally the ‘signaling proteins’ involved in sending messages between immune cells and cancer cells. In many malignancies, the expression of chemokines and their receptors is altered, leading to abnormal chemokine receptor signaling. So, fundamentally, chemokines are important regulators in the growth of many cancers as they play an important role in tumor progression, the formation of metastasis and chemotherapy resistance.

Researchers at the NIH (National Institutes of Health) have developed two novel technologies to specifically target the cells of interest in cancer. These technologies are based on the ability of chemokines to deliver antigens to the cells. The antigens cause an immune response, which allows the body to fight the tumor, instead of the cancer taking over the body!

One of these technologies is the use of a chemokine fused with toxic molecules (such as siRNA/miRNA), which specifically kills cells expressing the relevant chemokine receptors. This was further established through an experiment with mice, where immunization with chemokine-fused tumor antigen created an anti-tumor protective and therapeutic immunity. This technology can be employed for the treatment of cancerous diseases of human T-cells, such as T-cell lymphoma (white blood cell cancer starting in the lympatic system), melanoma (skin cancer) and T-cell leukemia (white blood cell cancer starting in the bone marrow).

This technology is effective and can be used to temporarily deplete the T-cells which are regulating or controlling the process. By stopping the process in its tracks, there is an ability to turn on the body’s anti-tumor immune response.

However, no methods are available to deliver siRNA/miRNA into the immune cells in the body. To circumvent this obstacle, a second technology called Chemo-ARP comes into play. Developed by the NIH, Chemo-ARP is a simple technique that inactivates the functions of immune cells by delivering oligonucleotides – this fundamentally slows down the tumor and metastasis. Chemo-ARP is a potent therapeutic strategy to modulate the body’s immune responses against cancer – allowing the body to mount its own attack against the cancer. By essentially helping the body ‘kill the tumor’ itself, we have a situation akin to our white blood cells fighting off an infection!

The use of chemokines reflect a multi-faceted approach:

  • Used to deliver tumor-associated antigens, thereby shifting the balance towards jump-starting the anti-tumor fighting power of the body’s immune system.
  • Can kill regulatory immune cells. This is important because these regulatory immune cells help the tumor to metastasize (spread).
  • Can control growth and metastasis of the cancer.

Chemokines are a simple and robust cancer strategy that can dramatically improve the outcome of the disease. This immunotherapeutic option (therapy to help the immune system do its job and destroy the tumor) can significantly bridge the gap between cancer and current treatment options.
